Up-regulation of the thioredoxin protein levels by IFN-γ. THP1 cells (5 × 105) were treated with media alone or with IFN-γ under serum-free conditions for the indicated time periods. The cells were then treated with brefeldin A (10 μg/ml) for 4 h prior to being harvested, after which they were permeabilized and stained with mouse monoclonal anti-thioredoxin antibodies and mouse IgG-FITC as described in the text. FACS analysis was performed to determine the intracellular protein levels based on their MFI values. Panel A: A representative FACS histogram. Panel B: ΔMFI values for thioredoxin levels.
